Enhancer experiment: CAT Assay,PCR,RT-PCR
Enhancer experiment description: Functional tests with the CAT reporter gene assays demonstrate that the human 59 HS5 LTR activates the cis-linked CAT gene and possesses enhancer and pro_x0002_moter activities in erythroid cells. We found an LTR retrotranspo_x0002_son belonging to the ERV-9 family of human endoge_x0002_nous retroviruses in the apparent 5* boundary area of the LCR. This ERV-9 LTR contains an unusual U3 en_x0002_hancer region composed of 14 tandem repeats with recurrent GATA, CACCC, and CCAAT motifs.

About Target gene

Target gene : HBB(CD113t-C,ECYT6,beta-globin)
Strong evidence: --
Less strong evidence: CAT Assay,RT-PCR
Target gene experiment description: We next determined whether the endogenous 59 HS5 LTR also exhibits enhancer and promoter activities and can activate the transcription of the downstream R region and the flanking genomic DNA in the b-globin LCR.The above RT-PCR results indicate that the endog_x0002_enous 59 HS5 LTR possesses apparent enhancer and promoter activities and is capable of promoting the transcription of the R and U5 regions in the LTR and of further downstream genomic DNA in the LCR.

About Function

Enhancer function : In both recombinant constructs and the endog_x0002_enous human genome, the LTR enhancer and promoter activate the transcription of cis-linked DNA preferentially in erythroid cells.
Enhancer function experiment: CAT Assay
