Enhancer experiment description: | To find the enhancers of the IFNγ gene,different 240-bp fragments of the gene were inserted between the GFP gene and the Alu tandem repeats in the C1-Alu14 plasmid.GFP expression in transfected cells was assessed using fluores_x0002_cence microscopy.We found that five different 240-bp fragments displayed strong enhancer activity.One enhancer was located in an upstream sequence of the IFNγ gene (−480~−720 bp,FUIFN3F3R);two enhancers (IFN4F4R and IFN6F6R) were located in intron 1;another two enhancers (IFN21F21R and IFN22F22R) were located in a noncoding region downstream of the IFNγ gene. The percentages of the GFP-positive cells for the five enhancers were 6.9%, 2.6%, 6.3%, 7.7%, and 4.9%,respectively.The ratio of the percentage of the GFP-positive cells induced by the inserting fragments compared with that of C1-Alu14 reflects enhancer activity. |
