Enhancer experiment description: Among seven constituents of miR-290-295 SE (E1-E7, Figure 4A), deletion of individual elements except for E6 led to substantial decrease (ranging from 50% to 80%) in de novo production of mature miRNAs in TT-FHAgo2 mESC background without concomitant changes in other miRNA production (Figures 4Band S5A). A 5-fold reduction in expression upon deletion of one constituent of a group of six indicates cooperative interactions.As expected from a decrease in these miRNAs, these deletions also suppressed expression of Ago2-inducible gene Lin28a, a regulator of ES cells (Figure 4B).
