Target gene experiment description: | In K562 cells, three copies of chromosome 11, which is the loca_x0002_tion of the _x0002_-globin locus, were detected in most nuclei by hybridizing a DNA probe corresponding to the centromeric _x0008_-satellite repeat region. RNA FISH performed on the same slides revealed that only two chromosomes actively transcribe ε-globin. |

To ascertain whether this is the case,we employed DNA/RNA FISH.Here, we studied the establishment and function of histone acetylation and transcription in noncoding sequences by using a model locus linking the ε-globin HS2 enhancer and the embryonic ε-globin gene in chromatin. An intact HS2 enhancer that recruits RNA Pol II is required for intergenic transcription and histone H3 acetylation and K4 methylation between the enhancer and target gene. |
