Enhancer experiment description: | To test whether β-catenin/TCF4 complexes were responsible for enhancer activity, we mutated the consensus TCF motif to CGCTGAT.In transfected cells, levels of luciferase expressed from the YAP reporter were _x0001_60% of the lev_x0002_els observed with the YAP (WT) reporter.To test whether the residual activity of the YAP reporter was due to a second β-catenin binding site, cells were treated with the GSK3_x0001_inhibitor BIO. BIO treatment increased both pGL3-YAP (WT) and pGL3-YAP reporter activities suggesting that the YAP enhancer element contains a second, unidentified β-catenin binding site. |

Target gene experiment description: | Because β-catenin is recruited to chromatin through interactions with TCF4, we next tested whether TCF4 bound to the YAP gene using ChIP assays.Here we demonstrate that β-catenin/TCF4 complexes bind a DNA enhancer element within the first intron of the YAP gene to drive YAP expression in CRC cells. As such, reducing β-catenin expression in CRC cells using shRNAs leads to decreased YAP mRNA and protein levels. |
