Microscopic analysis revealed that after induction of DNA damage, all five cell lines adopted a senescent phenotype with a large, flattened shape and multiple or enlarged nuclei, and enhanced the expression of SA-β-galactosidase.Quantification of BrdU-positive cells showed that DNA damaged cells strongly reduced the incorporation of BrdU,confirming the absence of S-phase activity. |
