Strong cellular staining for 5-bromo-4-chloro-3-indolyl-β-D-galactoside (X-gal) at pH 4, indicating physiological lysosomal β-galactosidase activity, was detected in control fibroblasts and in those bearing LMNA mutations or treated with PIs.LMNA mutations and PI treatment also induced a striking decrease in fibroblasts’ replicative capacity, measured in terms of bromodeoxyuridine (BrdU) incorporation in halfconfluent cells . |
