Although the TSA-induced reduction in the number of colonies formed 10 days after treatment did not reach statistical significance, TSA produced a mean decrease of 30, 73, and 53 %, respectively, in colony sizes. The results suggest that TSA inhibits the proliferation and survival of U87 GBM cells. In addition, TSA at 500 nM induced G2/M cell-cycle arrest.The presence of senescent cells was confirmed by a SA-β-galactosidase activity assay, in which senescent cells displayed morphological alterations similar to those observed in the NMA analysis. |
