Western blot： Inhibition of CDK4/6 activity with suboptimal doses of Palbociclib (0.05 μM) together with the inhibition of autophagy using doses of 0.5 μM o f Spautin-1 (SP-1), resulted in a reduction in AGS cell proliferation that was greater than the inhibition induced by Palbociclib-only treatment .SA-β-gal activity assay：the proportion of giant, multinucleated senescent cells that were positive for SA-β-Gal activity was higher in cells treated with both drugs .Western blot:Through immunofluorescence and western blotting, an increase in total p53 levels in AGS cells treated with Palbociclib was evident,p53-deficient cells display a reduced ability to induce senescence following Palbociclib treatment. |
