Our transcriptome analyses demonstrated that Treg-induced senescent CD8+ T cells induced significant alterations in genes involved in MAPK signaling pathways. We then confirmed the activation of MAPKs, including ERK1/2, p38 and JNK in na?ve CD4+ T cells treated with CD4+ CD25hiFoxP3+ Treg cells using Western blot analyses. We found that Treg-treated na?ve CD4+ T cells selectively activated ERK1/2 and p38, but not JNK, resulting in significantly enhanced phosphorylation of ERK1/2 and p38. We found that only the TLR8 ligands Poly-G3 and ssRNA40 significantly blocked the induction of responder T cell senescence induced by CD4+ CD25hiFoxP3+ Treg cells identified by SA-β-Gal expression . |
