| Cell name: | MTMEC |
| Experiment: | SA-β-gal activity assay//EdU assay |
| Description: | Overexpression of the miR-106bB25 cluster allowed cells to bypass doxorubicin-induced senescence and increased the capacity of MTMEC cells to generate doxorubicin-resistant cells. |
| Target gene: | EP300 |
| Official symbol(s): | EP300 |
| R-AG-Target gene: | Downregulation |
| Subcategory: | Unclear |
| Target gene experiment: | Luciferase reporter assay |
| Target gene description: | To experimentally confirm that EP300 mRNA was a target of the miR-106bB25 cluster, we used a luciferase reporter with the 30 -UTR of EP300 and transiently transfected either control or cells overexpressing the cluster or individual miRs. In all cases, luciferase expression from the plasmid containing the EP300 30 -UTR was lower in cells overexpressing both the miR-106bB25 cluster and its individual miRs.which overexpress the miR-106bB25 cluster, showed reduced EP300 and CDH1, but increased VIM(encoding vimentin, a mesenchymal intermediate filament protein activated during EMT4) mRNA levels. |
