| Target gene description: | Computational miRNA target analysis from a miRNA database demonstrated that miR-377 had high homology with a sequence in the 3′-UTR of human DNMT1 mRNA.DNMT1 mRNA and protein expression levels were significantly decreased by the miR-377 mimics and elevated by the miR-377 inhibitors.Of these three genes, p53 was further confirmed to be regulated by miR-377 using RT-qPCR and western blotting. Increased p53 mRNA,p53 and Rb expressions together with diminished phosphoryla- tion of Rb were induced by the miR-377 mimics and reverse changes were induced by the miR-377 inhibitors compared with that of the respective controls. |
