| Description: | We found that RNase-L-expressing cells did not display nuclear labeling, suggesting that ectopic expression of RNase-L inhibited proliferation. In contrast,the nuclei of cells with low,background staining for RNase-L expression were strongly labeled.RNase-L induction resulted that these cells exhibited an increased cytoplasmic :nuclear volume and vacuolization, and a flattened, multinucleate morphology.Counting of the stained cells revealed that 62% of RNase-L-transduced WI38 HDFs were β-gal positive as compared to only 18% in the vector control cells. Replicative senescence was accelerated by nearly three weeks in the RNase-L WI38 as compared to vector control cells. |
