| Description: | The MTT assay showed that MED12 KO clones proliferated significantly slower from day 2 compared with the MED12 WT or MED12 reconstitution single clones. This phenomenon was further confirmed by a colony formation assay. We found that MED12 knockout caused multinucleation in PC9 cells and restoring MED12 expression in MED12 KO cells was sufficient to avert multinucleation. A similar phenomenon was observed in SPC-A1 cells. Next, we investigated the cell fate of multinucleated cells and found that they exhibited senescence. |
