| Description: | Growth of EZH2 siRNA cells was significantly decreased compared to control-transfected cells. EZH2 siRNA cells displayed a flattened, enlarged morphology and expressed SA-β-Gal. In addition to SA-β-Gal expression, additional senescence markers following extended EZH2 depletion (6 days) include SAHFs and the heterochromatin-associated H3K9me3 modification as well as an increase in cell size, as evaluated by tubulin immunofluorescence. Flow cytometric analysis demonstrated that the proportion of cells in the S phase decreased in EZH2 siRNA cells with a concomitant increase of cells in the G1 phase, a significant difference in all 3 cell lines tested. In SK-Mel-173 cells, we also noticed a slight increase in the proportion of cells in the G2 phase. |
